- Breathing in air containing particulate matter (PM) derived from human activity (i.e., traffic, power plants, and agriculture) has a detrimental impact on human health and is associated with many prevalent cardiopulmonary diseases (3, 6). Longterm exposure to PM is linked to a decline in lung function and the development and/or progression of respiratory diseases including chronic obstructive pulmonary disease (COPD), asthma, and idiopathic pulmonary fibrosis (IPF) (6, 31, 37a). Ambient PM is a complex mixture of solid and liquid particles containing soluble transition metals, organic compounds, and endotoxins that vary in composition and size, depending on the location, time, and weather when sampled. PM with an aerodynamic diameter of less than 10 m is respirable into the airways, while PM of diameter less than 2.5 m can penetrate the small airways and lung parenchyma and pose a greater health risk. Accumulating data suggest that the primary underlying pathogenic mechanism of PM-evoked lung injury and disease involves the metal-catalyzed generation of reactive oxygen species (ROS) and subsequent DNA and mitochondrial damage (24, 28, 35). However, there remain large gaps in our knowledge of the mechanisms and cellular processes by which particulate air pollution contributes to the pathobiology of chronic lung disease.
