- When plant cells or tissues are cultured in vitro with suitable culture media they can be directed into different developmental pathways. This capacity has been demonstrated in numerous species with many different cell and tissue types and different culture media. Plants are more readily directed into regeneration than animal cells but this capacity to genetically reprogram in vitro under suitable conditions is characteristic of eukaryotic cells. In plants, the capacity to reprogram cell fate using plant hormones, the auxins and cytokinins, was demonstrated many years ago (Skoog and Miller 1957). In this Chapter three types of development reprogramming are considered: somatic embryogenesis (SE), root organogenesis and shoot organogenesis. Somatic embryogenesis involves the formation of both shoot and root meristems and the capacity to produce a normal flowering plant (Steward et al. 1958). With root organogenesis a root meristem develops and differentiates into a root with a capacity to produce lateral roots; and with shoot organogenesis there is the development of a shoot apical meristem with the capacity to differentiate into the tissues of the stem, its buds and reproductive tissues (Skoog and Miller 1957). Callus is a form of differentiation and this is addressed in the SE and organogenesis context. A major focus in this Chapter is on the genetic reprogramming to form stem cells in vitro from which the embryos or organs are formed or reprogramming from pre- existing stem cells (Verdeil et al. 2007, Wang et al. 2011); with particular emphasis on SE.
